GENETIC DIVERSITY AND RELATEDNESS AMONG ACCESSIONS OF RAMBUTAN (NEPHELIUMLAPPACEUM)

ABSTRACT
Randomly amplified polymorphic DNA (RAPD) markers were successfully employed to reveal genetic diversity and genetic relatedness among 211 Nephelium lappaceum accessions planted at the MARDI-IPGRI rambutan field genebank, Kemaman, Terengganu. A total of 270 clear, consistent and reproducible fragments were generated for Nephelium accessions by using eleven single random oligonucleotide primers. Genetic variation is high among the N. lappaceum accessions, with an average degree of polymorphism of 98.45% and mean Shannon index, H0 = 6.00. The RAPD technique was able to differentiate all the N. lappaceum accessions studied as well as two TV. ramboutan-ake accessions (i.e. PR155 and PR189) found in the same field genebank. The Jaccard index of genetic similarity ranged from 0.232 to 0.900 among the accessions, with a mean value of 0.444.
ABSTRAK
Penanda DNA polimorfik teramplifikasi rawak (RAPD) telah digunakan untuk melihat kepelbagaian genetik dan hubungan genetik antara 211 aksesi Nephelium lappaceum di bank gen rambutan MARDI-IPGRI, Kemaman, Terengganu. Sejumlah 270 jalur yangjelas, mantap dan boleh diulangi telah terhasil bagi aksesi Nephelium tersebut dengan menggunakan sebelas pencetus oligonukleotida rawak tunggal. Variasi genetik adalah tinggi antara aksesi-aksesi N. lappaceum, dengan min darjah polimorfisme 98.45% dan min indeks Shannon, H0 = 6.00. Teknik RAPD berjaya membezakan kesemua aksesi N. lappaceum dan juga dua aksesi N. ramboutan-ake (iaitu PR155 dan PR189) yang terdapat dalam bank gen yang sama. Indeks Jaccard yang memaparkan darjah kesamaan genetik didapati berjulat antara 0.232 dan 0.900 di kalangan aksesi-aksesi yang dikaji dengan nilai min 0.444.
