THE EFFECT OF SOLUBLE CARBON SOURCES ON THE

PRODUCTION OF CHITOSAN BY A NEW ISOLATE OF

RHIZOPUS OLIGOSPORUS

ABSTRACT

Phis research was undertaken to study the effect of different soluble carbon sources on the production of chitosan by Rhizopus oligosporus. The glucose content in the TVB medium was substituted with D-fructose, D-sucrose, D-maltose or D-lactose at concentrations of 1 to 3% (W/V). From the four carbon sources used, D-fructose produced the highest biomass of 4.35gm/250ml of growth medium, with the highest yield of chitosan obtained when 2% D-fructose medium was used (Yp/s=0.42). There was no significant difference in the production of chitosan between 2% D-fructose and 2% D-glucose medium (a=0.05). Results on the effect of pH and temperature showed that pH 3.0 and the temperature of 30°C (4.45gm biomass/250ml medium) was the best for maximum yield of chitosan. The maximum production of chitosan obtained under optimum conditions was 0.813gm /4.35gm biomass/250ml medium (Yp/, = 0.19) obtained in a 2% fructose medium.

ABSTRAK

Kajian in dijalankan untuk melihat kesan sumber karbon berlainan terhadap penghasilan kitosan oleh Rhizopus oligosporus. Kandungan glukosa dalam medium TVB telah digantikan dengan D-fruktosa, D-sukrosa, D-maltosa atau D-laktosa pada kepekatan 1 hingga 3% (W/V). Daripada keempat-empat sumber karbon yang digunakan D-fruktosa menghasilkan biojisim tertinggi iaitu sebanyak 4.35gm/250ml media pertumbuhan, dengan penghasilan kitosan tertinggi diperolehi dengan menggunakan 2% D-fruktosa (Yp/s=0.42). Tiada perbezaan yang signifikan (a=0.05) diperhatikan dalam penghasilan kitosan apabila 2% D-fruktosa dan 2% D-glukosa digunakan. Hasil yang diperolehi dalam penentuan kesan pH dan suhu terhadap penghasilan kitosan menunjukkan bahawa pH 3.0 dan suhu 30°C menyumbang kepada penghasilan kitosan tertinggi. Penghasilan kitosan maksimum yang diperolehi dalam media pertumbuhan 2% fruktosa dalam keadaan pertumbuhan optimum adalah 0.813gm /4.35gm biojisim/250ml media (Yp/x =0.19).
