KEPELBAGAIAN MIKROORGANISMA TANAH DARIPADA TAMAN PAKU PAKIS, UNIVERSITI KEBANGSAAN MALAYSIA

ABSTRACT

In this study, six bacterial and seven fungal isolates from the soil of the Fernarium (Taman Paku Pakis) at Universiti Kebangsaan Malaysia were successfully isolated. Identification based on morphology, microscopy, biochemistry and molecular techniques revealed that three bacterial species were identified as Staphylococcus saprophyticus (RA, RD and RF), Bordetella avium (RB) and Bacillus cereus (RE). Identification of microorganisms using the 16S rRNA nucleotide sequence comparison showed that similarity between the bacterial species was between 84-94%. For fungal identification and isolation, six of the seven colonies isolated were identified to the genus level. Two of the isolates were from the genus Aspergillus (RK1 dan RK3), two from genus Paecilomyces (RK4 and RK6) and one isolate each of the genus Gliocladium (RK5) and Acremonium (RK7). RK2 did not show any spore production but showed septate mycelium. The fungi isolated from this study were classified in the same class namely Deuteromycetes and order Moniliales.


ABSTRAK

Dalam kajian ini, sebanyak enam pencilan bakteria dan tujuh pencilan kulat berjaya dipencilkan daripada sampel tanah di Taman Paku Pakis, Universiti Kebangsaan Malaysia. Melalui kaedah pengenalpastian secara morfologi, mikroskopi, biokimia dan molekul, tiga spesies bakteria telah dikenalpasti iaitu Staphylococcus saprophyticus (RA, RD and RF), Bordetella avium (RB) dan Bacillus cereus (RE). Pengenalpastian mikroorganisma melalui perbandingan jujukan nukleotida 16S rRNA mengesahkan kehadiran spesies bakteria diatas dengan peratus kesamaan nukleotida antara 84-94%. Bagi pengenalpastian pencilan kulat secara morfologi pula, daripada tujuh pencilan, hanya enam daripadanya sahaja dapat dikenalpasti sehingga peringkat genus iaitu dua pencilan kulat daripada genus Aspergillus (RK1 dan RK3), dua pencilan kulat dari genus Paecilomyces (RK4 dan RK6) dan satu pencilan kulat bagi setiap genus Gliocladium (RK5) dan Acremonium (RK7). RK2 tidak menunjukkan pembentukan spora yang jelas tetapi mempunyai miselium bersepta. Kesemua pencilan kulat tergolong dalam kelas yang sama iaitu Deuteromycetes dan order Moniliales.
