ANALYSIS OF O-PHTHALDIALDEHYDE PRECOLUMN DERIVATIZED PHYSIOLOGICAL AMINO ACIDS

ABSTRACT
This paper describes the application of a new reversed-phase high-performance liquid chromatographic system to the analysis of o-phthaldialdehyde precolumn derivatized free amino acids in plasma and brain tissue. Good separation of 23 individual amino acids was achieved within 45 min. The coefficient of variation for retention times for each amino acid, except ASP and GLU, was around the range of 1 %. The variability of peak areas for most amino acids were less than 5 % for within-day and between-day determinations. Good linearity was obtained for each standard amino acid over the concentration range of 35 to 100 pmol. These results suggest that the present system is flexible, reliable and feasible for routine analysis of free amino acids.
ABSTRAK
Kertas ini menerangkan penggunaan sistem baru kromatografi cecair fasa terbalik untuk analisis asid amino bebas terderivasi >phthaldialdehyde dalam plasma dan tisu otak. Pengasingan yang baik 23 asid amino individu tercapai dalam 45 min. Koefisien variasi masa pengekalan setiap asid amino, kecuali ASP dan GLU, berbanjaran 1 %. Variabiliti kawasan puncak cebanyakan asid amino adalah kurang dari 5 % antara dalam-hari dan antara-hari. Keselanjaran yang baik diperolehi intuk setiap asid amino dalam banjaran kepekatan 35 hingga 100 pmol. Keputusan ini menunjukkan sistem ini adalah leksibel, boleh dipercayai dan boleh dilaksanakan untuk analisis rutin asid amino bebas.
