POPULATION GENETIC STRUCTURE OF BAUNG, MYSTUS NEMURUS (C&V), IN MALAYSIA

ABSTRACT
Wild and hatchery samples of Mystus nemurus (C&V), from seven populations were analysed using starch gel electrophoresis. Samples from muscles, liver, heart and kidney were used to examine genetic variation from thirteen enzyme systems and a sarcoplasmic protein. These populations were from Pulau Pinang, Kelantan, Selangor, Terengganu, Melaka, and Johor in the north, northeast, central and south of West Malaysia and from Sarawak in East Malaysia. Sixteen of 25 loci examined (64%) were found to be polymorphic (at P= 0.95) namely AAT-1*, AAT-2*, ACP-1*, ACP-2*, EST-1*, EST-2*, GPI-1*. IDH-1*, IDH-2*, LDH-1*, MDH-1*, MDH-2*, ME-1*, PGM-1*, SP-1* and 6-PGD*. Gene frequencies of the M. nemurus populations were largely in Hardy-Weinberg equilibrium with high genetic variability and observed heterozygosities ranging from 0.004 to 0.104. Dendrogram derived from the genetic distances based on the allele frequencies showed low genetic similarity among populations of M. nemurus except for Terengganu population, indicating a substantial level of differentiation from the rest of the seven populations.
ABSTRAK
Sampel Mystus nemurus daripada 7 populasi liar dan ternakan dianalisis dengan menggunakan elektrotbresis gel kanji. Tisu otot, hati, hepa dan ginjal digunakan untuk menentukan variasi genetik berdasarkan kepada tiga belas sistem enzim dan protein sarkoplasma. Populasi-populasi ini didapati dari Pulau Pinang, Kelantan, Selangor, Terengganu, Melaka dan lohor yang masing-masing terletak di utara, timur laut, tengah dan selatan Malaysia Barat, dan dari Sarawak di Malaysia Timur. Enam belas daripada 25 lokus yang dikaji adalah polimorfik (pada P=0.95) iaitu AAT-1*, AAT-2*, ACP-1*, ACP-2*, EST-1*, EST-2*, GP1-1*, IDH-1*, IDH-2*, LDH-1*, MDH-1*. MDH-2*, ME-1*, PGM-1*, SP-1* and 6-PGD*. Frekuensi gen bagi populasi M. nemurus ini adalah sebahagian besarnya mengikut keseimbangan Hardy-Weinberg dengan variabiliti genetik dan heterozigositi tercerap berjulat di antara 0.004 ke 0.104. Dendrogram yang diperolehi daripada jarak genetik berdasarkan frekuensi alel menunjukkan persamaan genetik yang rendah di kalangan M. nemurus kecuali populasi Terengganu, yang menunjukkan perbezaan daripada 7 populasi yang dikaji.
